建立了三倍体毛白杨遗传转化系统 :以茎段为外植体 ,基本培养基是MS +IBA 0 .2 5mg/L +ZT 0 .2 5mg/L +TDZ 0 .0 5mg/L ,农杆菌菌株用EHA10 5 ,菌液浓度OD6 0 0 值为 0 .3 ,浸染 3 0min ,在共培培养基中加入乙酰丁香酮 2 0 0 μmol/L .

We selected stems as explants to establish Agrobacterium-mediated high frenquency transformation system for Populus tomentosa:the primary medium was MS+IBA 0.25 mg/L+ZT 0.25 mg/L+TDZ 0.05 mg/L,and the stems were floated in liquid medium containing the A. tumefacenstrain EHA105 for 30 minutes.
