摘要 研究了固相萃取 -高效液相色谱测定烟草样品中的植物色素的方法。烟草样品中的植物色素用 Sep- Park- C1 8固相萃取小柱预分离和富集 ,以 C1 8为固定相 ,(1+1)甲醇异丙醇溶液 -水梯度为流动相 ,各组分均在最大波长下提取色谱图计算峰面积定量。方法标准回收率为 92 %~ 10 1%,RSD为 1.2 4%~ 2 .31%。用该方法研究了烤烟新鲜烟叶烘烤过程中色素变化 ,结果令人满意。 A high performance liquid chromatography (HPLC) method for the determination of tobacco leaf pigments was studied. Tobacco leaf pigments were extracted from tobacco leaf with 90 acetone by oscillating and were enriched by solid phase extraction with Sep-Park-C 18 cartridge. Chlorophyll-a, Chlorophyll-b, β-Carotene, lutein, neoxanthin, violaxanthin, pheophorbide and other tobacco leaf pigments were separated on a Nova-Pak-C 18 chromatogram column (3.9×150mm), with methanol-isopropyl alcohol-water gradient elution as mobile phase at a flow-rate of 0.5mL/min. The recoveries of tobacco leaf pigments are 92~101, and relative standard deviations are 1.24~2.31. This method can be applied to determine pigments in tobacco leaf in the course of curing with satisfactory result.
