摘要
目的 :制备鼠抗人血管抑素 (angiostatin)的单克隆抗体 ,为进一步研究血管抑素的抗肿瘤作用和临床应用打下基础。方法 :利用杂交瘤技术以血管抑素免疫BALB/C小鼠制备 3株能够稳定分泌抗血管抑素抗体的杂交瘤 ,并对上清进行ELISA、双向免疫扩散等鉴定。结果 :该抗体的类别为IgG1,并能特异性识别血管抑素 ,与细胞因子rhIL - 2、rhTNF -α、rhIFN -α及血清蛋白无交叉反应。结论 :经初步鉴定获得
AIM:To further study the anti-tumor effect of angiostatin, an anti-human angiostatin monoclonal antibody was prepared and identified.METHODS:The hybrodoma techniques were used. The BALB/C mice were immunized with angiostatin. The supernatant of cell culture were collected and screened by ELISA and double immunodiffusion.RESULTS: There cell lines which steadily secreted the anti-angiostatin monoclonal antibody were identified by ELISA and double immunodiffusion. The antibody was IgG1 and specifically recognized angiostatin without crossing reactions to rhIL-2, rhTNF-α, rhIFN-α and serum proteins.CONCLUSION: The antibodies secreted by three hybridoma cell lines identified by several methods were specific antibodies of angiostatin.
