裸小鼠可移植人脑胶质瘤中VEGF、vWF的表达和血管形成关系的初步研究

摘要 目的 探讨血管内皮细胞生长因子 (VEGF)、温韦伯因子 (vWF)在裸小鼠可移植人脑胶质瘤不同生长阶段的表达规律以及对血管形成的作用。方法 建立移植瘤动物模型 ;用组化及免疫组化的方法处理 30例移植瘤、6例对照组移植瘤和 6例正常脑组织石蜡标本 ;统计分析病理指数、微血管密度、坏死指数和瘤重等指标。结果 移植瘤中VEGF的表达阳性率为 10 0 % ,与坏死指数和微血管密度呈显著正相关 (r为 0 .5 2 4、0 .4 96 ,P <0 .0 1) ,与瘤重不相关。结论 VEGF在移植性人脑胶质瘤的血管形成及肿瘤的发展、侵袭和转移中具有重要作用。VEGF和微血管密度可作为胶质瘤恶性生物学行为的指标之一。 Objective To investigate the expression and role of vascular endothelial growth factor (VEGF) and von Willebrand Factor (vWF) on the angiogenesis of transplanted human glioma in different growth period. Methods Using immunohistochemistry and histochemistry to analysis transplanted human glioma( n =30),controlled transplanted glioma( n =6)and normal human brain tissue( n =6) paraffin-embedded samples. The data of pathological index, micro-vascular density (MVD), necrosis index and glioma weight were analyzed statistically. Results The expression rate of VEGF was 100% in transplanted glioma. The VEGF was closely correlated with necrosis index and MVD ( r =0.524 0.496, P <0.01) but not with glioma weight. Conclusions VEGF plays an important role in angiogenesis, growth and metastasis of transplanted glioma. MVD and VEGF can act as malignant biological behavior marker of glioma.
