电离辐射对人肺腺癌细胞株A549体外表达VEGF的影响 被引量:1

摘要 目的 探讨电离辐射对人肺腺癌细胞体外表达血管内皮生长因子 (VEGF)的影响。方法 人肺腺癌细胞株A5 4 9,置RPMI 16 4 0培养液培养 ,分别接受 1、2、4、8Gy电离辐射 ,继续培养细胞 ,2 4、4 8h后收集细胞上清液 ,双抗体夹心ELISA法测定VEGF表达水平。结果 与阴性处理细胞比较 ,人肺腺癌细胞株A5 4 9体外接受不同剂量辐照后 ,VEGF表达明显增高 ,且具有剂量依赖性和时程依赖性。结论 电离辐射能诱导肺腺癌细胞VEGF高表达 ,这可能是临床肿瘤抗辐射的机制之一。 Objective To investigate the effects of ionizing radiation (IR) on vascular endothelial growth factor (VEGF) expression in human lung adenocarcinoma cells.Methods A549 human lung adenocarcinoma cells were maintained in RPMI 1640 medium and exposed to IR in incremental doses of 1, 2, 4 and 8 Gy. These cells continued to be cultured in the same medium for 24 and 48 hours after exposure, and the media were harvested for measurement of VEGF protein levels by ELISA. Results Exposure of A549 cells to IR induced significant upregulation of VEGF expression. VEGF levels exhibited both an IR dose- and time-dependent increase in comparison with the control unirradiated cells.Conclusion IR-mediated induction of VEGF in lung adenocarcinoma cells may contribute to clinically-related tumor radio-resistance.
