醋酸亮丙瑞林一级结构的电喷雾质谱分析

Determination of First-order Structure of Leuprorelin Acetate by Electrospray Ionization Mass Spectrum

目的 :醋酸亮丙瑞林的分子量及其一级结构的测定。方法 :应用电喷雾质谱法 (ESI/MS)及源内碰撞诱导解离技术对两端封闭的九肽药物醋酸亮丙瑞林进行测定 ,根据其典型碎片离子确证其氨基酸序列及分子量。结果 :测得亮丙瑞林准分子离子峰 (M +H) + m/z 12 0 9 7及 (M +Na) + m/z 12 31 6 ,(M +2H) 2 + m/z 6 0 5 4及 (M +H +Na) 2 + m/z 6 16 5 ,表明亮丙瑞林的分子量为 12 0 8 7,与理论值一致。适当增加碎裂器电压 ,得到一系列典型的y及b系列碎片离子 ,证实亮丙瑞林的氨基酸序列为pE H W S Y L L R PNHEt。结论 :亮丙瑞林的分子量及一级结构与理论值相符。

AIM:To determinate the molecular weight and the first-order structure of leuprorelin acetate. METHOD:The molecular weight and amino acid sequence of leuprorelin were determined by electrospray ionization mass spectrametry.A series of typical fragment ions were obtained by in-source collision-induced dissociation(CID). RESULT:The quasi-molecular ions were 1209.7 (M+H) + and 1231.6 (M+Na) + . The double-charge ions were 605.4 (M+2H) 2+ and 616.5(M+H+Na) 2+ . The y and b series of fragment ions were obtained by adjusting the fragmentor voltage. CONCLUSION:The molecular weight and first-order structure of leuprorelin acetate were confirmed.
