摘要
目的:研究9-顺-维甲酸对非小细胞肺癌A549、PG、SPC-A1的细胞周期蛋白D1(cyclinD1)、细胞周期蛋白依赖性蛋白激酶4(Cdk4)表达的影响,探讨其抑制非小细胞肺癌生长与影响CyclinD1、Cdk4表达的相关性。方法:采用半定量逆转录PCR方法,分析细胞周期因子CyclinD1、Cdk4表达的变化。结果:9-顺-维甲酸既可以降低PG、SPC-A1的CyclinD1表达(P<0.01),又可以降低PG、A549的Cdk4表达(P<0.01)。结论:9-顺-维甲酸对非小细胞肺癌生长的抑制作用,与其调控细胞周期因子CyclinD1、Cdk4的表达密切相关。
Objective:To study the effects of9-cis-retinoic acid(9-cis-RA)on the expression of CyclinD1and Cdk4in non-small-cell lung cancer PG,A 549 and SPC-A1,and to investigate the correla-tion between inhibition of cell growth by9-cis-RA and interference with the expression of CyclinD1and Cdk4in non-small-cell lung cancers.Method s:RT-PCR was used to analyze the changes in the expression of CyclinD1and Cdk4of PG,A 549 and SPC-A1.Results:9-cis-RA can not only decrease the transcription of CyclinD1of PG and SPC-A1(P<0.01),but also decrease the transcription of Cdk4of PG and A 549 (P<0.01).Conclu sion:The growth-inhibitory effect of9-cis-RA on non-small-cell lung cancers was tightly correlated with its mediation of the transcription of CyclinD1and Cdk4.
