摘要
目的细胞外基质成分与腺样囊性癌(Adenoidcysticcarcinoma,ACC)特殊病理学结构的形成和侵袭等生物学表型关系密切。本研究观察来源于人腺样囊性癌的细胞株ACC2合成细胞外基质成分I、II型胶原蛋白的过程,并探讨其与ACC囊腔形成的关系。方法应用间接免疫荧光组织化学方法观察蛋白质的动态合成过程,并以RT-PCR法检测相应mRNA的表达。结果ACC2有I型胶原mRNA表达,免疫荧光观察到I型胶原合成和分泌的动态过程。同时在基因和蛋白水平ACC2都无II型胶原的合成。结论I型胶原是ACC基质的重要成分,与其囊腔形成关系密切。II型胶原不参与囊腔的形成。
Objective To observe the biosynthese s of collagen type I,II in ACC2cells,established from adenoid cystic carcinoma(ACC )of the salivary gland.Methods RT-PCR and immunofluorescence were employed to in vestigate gene and protein levels in ACC2.Results The production of collagen I w as detected in ACC2.ACC2didn't synthesized colla-gen II.Conclusion Collagen I i s involved in the formation of pseudocysts structures of ACC.
