摘要
目的研究加热对人舌癌Tca8113细胞尿激酶型纤溶酶原激活物(urokinase-typeplasminogenactivator,UPA)mRNA表达的影响。方法根据不同加热温度分为37℃、40min;40℃、40min;43℃、40min;45℃、40min4组。按上述分组在不同加热剂量下加热人舌癌Tca8113细胞,采用原位杂交方法检测Tca8113细胞UPAmRNA的表达,并用Spot及IPP图像采集分析系统处理原位杂交染色结果。结果Tca8113细胞UPAmRNA阳性表达呈紫蓝色,定位于细胞浆。40℃、43℃组UPAmRNA染色的光密度值均小于37℃组(P<0.05);45℃组与37℃组相比,UPAmRNA染色的光密度值统计学上无显著性差异。结论40℃、40min和43℃、40min加热,可抑制人舌癌Tca8113细胞UPAmRNA的表达。提示:口腔癌热疗的常用加热剂量43℃、40min加热人舌癌Tca8113细胞后,不但没有促进Tca8113细胞UPAmRNA的表达,而且还抑制其表达。
Objective To observe the expression of urokin as e-type plasminogen activator(UPA)mRNA in human tongue squamous cell carcinoma ce ll line(Tca8113)at different dosages of heating.Methods According to the differe nt temperature of heating,the test was divided into four groups as follow:(1)37 ℃,40min heating group;(2)40℃,40min heating group;(3)43℃,40min heating group;( 4)45℃,40min heating group.Tca8113cells were heated and the expres-sion of UPA mRNA was examined using in situ hybridization(ISH).The staining results of ISH w ere analysed using Spot and IPP image analysis system.Results The expression of UPA mRNA was the positive staining in the cytoplasm of Tca8113cells.The staining intensity of UPA mRNA at40℃,43℃,45℃was respectively lower than that at37℃.T here were statistically significant differences(P<0.05)between40℃,43℃and37℃;w hile no significant differences(P>0.05)between at45℃and at37℃.Conclusions Heat ing can inhibit the expression of UPA mRNA of human tongue squamous cell carcino ma cells(Tca8113)at the heating dosage40℃,40min and43℃,40min,43℃.40min is the usual dosage of o-ral squamous cell carcinoma local hyperthermia in clinic.At this heating dosage,heating not only doesn't promote the ex-pression of UPA mR NA of human tongue squamous cell carcinoma cells(Tca8113),but also inhibits it.
