摘要
目的 研究特异性抗内毒素鸡蛋黄免疫球蛋白理化性质 ,探索一种防治肠源性感染 ,治疗内毒素血症的新途径。方法 用大肠杆菌J5株 (菌体表面类脂A裸露 )作抗原免疫 2 5周龄Roman鸡 ,制备特异性抗内毒素IgY ,用酶联免疫吸附技术检测其效价及对热、酸、碱和酶的稳定性。结果 IgY抗体效价为 1∶2 5 6 0 0 ,能耐受巴氏消毒 ,对酸、碱、酶有很好的稳定性。结论 用大肠杆菌J5株免疫鸡制备的IgY效价高 ,理化性质稳定 。
Objective To study physicochemical characteristics of specific chicken egg yolk immunoglobulin (IgY) against lipopolysaccharide (LPS) in order to explore a new way of prevention and cure of enterogenic infection and endotoximia.Methods Roman hens, 25 weeks old, were immunized with Escherichia coli J5 to produce IgY against LPS.The stability of IgY against heat, pH and enzyme was detected by enzyme labeling linked immunoadsorbent assay(ELISA).Result Minimal positive titer of IgY was 1∶25 600 and it was shown to be fairly stable against heat, pH and enzyme.Conclusion IgY against LPS produced by immunizing Roman hens with Escherichia coli J5 had high titer and stable physico-chemical characteristics, which would be fit to be taken orally.
