摘要
为探寻南瓜中对正常及糖尿病模型小鼠血糖有影响的有效成份,本实验采用新的分离工艺从南瓜粉中提取得到南瓜粗多糖(PP),用离子交换纤维素柱层析分析获得3个组分,收集的主导组分经过Sephadex G-100柱分级,以小白鼠血糖值作为筛选活性成分的指标,收集有活性的组分(PP-CG)经SephadexG-200证实为单一峰.气相色谱分析其单糖组成为葡萄糖、半乳糖、阿拉伯糖及鼠李糖.高效液相色谱证明其为杂多糖,分子质量为1.16×105u.
In Order to investigate the effect of unknown active Component from pumpkin on normal mice and alloxan induced diabetic mice,the experiment adopted a kind of new technology to extract and gain the pumpkin polysaccharide from pumpkin powder.This crude polysaccharide was fractionated by ion-exchange chromatography on DEAE and the main group-element puri- fied by gel filtration on Sephadex G-100.Its active part(PP-CG)was further purified on Sephadex G-200,and to be proved a homogeneous peak.By gas chromatography analysis,PP- CG composed of glucose,galactose,arabinose and rhomnose and its molecular weight is 1.16× 10u by HPLC.
