实验性脑出血大鼠脑组织Bcl-2,Bax蛋白表达特点及中药干预 被引量:6

摘要 目的 :从Bcl 2 ,Bax蛋白表达的时相、部位、量的变化特点 ,探讨其与脑出血损伤之间的关系及中药脑溢安对出血损伤脑组织的保护作用机制。方法 :Rosenberg方法复制大鼠脑出血模型 ,采用免疫组化法检测脑溢安组与各对照组在损伤的不同时相与病理分区中Bcl 2 ,Bax的阳性细胞 ,观察分析两种阳性细胞的变化特征。结果 :出血区可见Bax的表达 ,其阳性神经元形态变化较大 ,损伤较重 ;缺血反应区可见Bcl 2表达 ,其阳性神经元形态正常 ;出血边缘区可见Bax及Bcl 2的表达 ,在 2d ,4d ,7d脑溢安组与模型组比较Bcl 2表达增强 (均P <0 .0 1 ) ;在 1 2h ,1d ,2d ,4d ,7dBax表达降低 (均P <0 .0 1 )。结论 :大鼠脑出血后Bcl 2 ,Bax表达与脑组织损伤程度、时相、部位有关 ;脑溢安能上调Bcl Objective To investigate the characteristics of Bcl 2 and Bax expressions in rat brain after experimental intracerebral hemorrhagic (ICH), and to determine the protective mechanisms of the Traditional Chinese medicine complex, naoyian granule (NYA). Methods ICH models were induced according to Rosenberg's method. The rats were divided into the model group, the NYA treatment group and the sham group. Bcl 2 and Bax expressions in the rat brain after ICH were detected by immunohistochemistry method. Results Bax expressed in the hemorrhagic core, and the immunoreactive neurons were seriously injuried and the shape varied. Bcl 2 expressed in the penumbra with the normal shape of the immunoreactive neurons. Both Bax and Bcl 2 expressed simultaneously in the marginal region of the cerebral hemorrhagic core. Bcl 2 expression in the rat brain reached the peak at the 2 nd day and maintained at a relatively high level until the 7 th day, and NYA could increase it. Bax expression peaked at the 1 st day, and NYA could down regulate it. Conclusion The expressions of Bcl 2 and Bax after the cerebral hemorrhage are related to the degree of injury,the time, and the location of the brain tissues.NYA can up regulate the Bcl 2 expression and down regulate the Bax expression , which may be one of its neuroprotective mechanisms.
