大鼠脑出血后AQP4mRNA的表达与Ca^(2+)关系的探讨 被引量:23

摘要 目的 探讨AQP4mRNA的表达与Ca2 + 浓度在大鼠脑出血后脑水肿中的作用机制。方法 采用胶原酶复制大鼠脑出血模型 ,通过半定量逆转录聚合酶链反应 (RT -PCR)检测AQP4mRNA的表达 ,Fura - 2 /AM荧光法测定水肿区脑细胞内Ca2 + 浓度。结果 脑出血后血肿周围脑组织AQP4mRNA的表达和Ca2 + 浓度均明显增高。结论 脑出血后AQP4的表达增高 ,介导Ca2 + 内流增加 ,AQP4和Ca2 + 可能共同参与脑出血后脑水肿的形成。 Objective To investigate the expression of AQP4mRNA and the changes of Ca 2+ concentration during brain edema after intracerebral hemorrhage(ICH) in rats.Methods The model of ICH was made by infusing collagenase into rat caudate nucleuses.The expression of AQP4mRNA was assessed by RT-PCR. Fura-2/AM fluoremetry technique was used to determine the cellular free calcium concentration of brain tissue.Results AQP4mRNA expression and Ca 2+ concentration of perihemotoma tissue had significant increase after intracerebral hemorrhage. Conclusion The increased expression of AQP4mRNA after intracerebral hemorrhage participated in the increasing of focal concentration of cytosolic free calcium,which indicated that the AQP4 and Ca 2+ might correlate significantly with the brain edema formation.
