摘要
目的研究水杨酸钠对肾性高血压大鼠畸变产物耳声发射的影响。方法将造模4周后模型成功大鼠48只(包括假手术组12只)随机分成A、B、C、D4组(每组12只)进行实验。动物处死前检测DPOAE,并进行耳蜗组织SOD活性及MDA的含量和血浆ET含量的测定。结果①血浆ET含量D组与B组比较有明显降低(P<0.01),C组与B组比较无明显差异;②A组、D组、C组SOD活性明显高于B组(P<0.01);③A组、D组、C组耳蜗组织MDA平均含量明显低于B组(P<0.05);④在70dBSPL的高刺激强度下A、B、C、D四组各频率的DROAE检出率均为100%。在低刺激强度(49dBSPL)下,假手术组检出率为100%,而B、C、D组检出率则明显降低,与A组相比较在各频率DPOAE检出率有显著(P<0.05)或极显著(P<0.01)性差异。⑤B组(2K1C组)和C组较A组在各频率DROAE幅值均有所下降,但二者间无显著性差异(P>0.05)。而C组(水杨酸钠给药组)与A组比较在各频率DPOAE检出率的幅值均有极明显下降(P<0.01),尤其以高频为显著。③A、B、C、D4组对各频率DPOAE的潜伏期均无显著性差异(P>0.05)。结论①水杨酸钠可能通过改变内耳微循环而影响肾性高血压大鼠畸变产物耳声发射②水杨酸钠可以影响耳蜗组织自由基的代谢,并有利于氧自由基的清除,水杨酸钠对耳蜗组织氧自由基的清除并不改变其对DPOAE的影响。
Objective To study the effects of sodium salicylate on the distortion product otoacoustic emission(DPOAE) in rats with renovascular hypertension. Methods Forty-Eight rats with renovascular hypertension (including the 12 rats of shame group) was divided into four groups (groupA, B, C and D). The animals were determined by DPOAE before experiment and then sacrificed. The ET in plasma was measured by radioimmunoassay. MDA,SOD in cochlear tissues were measured by colorimetric method. Results ①Comparing with group B(group2K1C) ,the ET in plasma of group D was decreased markedly (P < 0. 01). ②The activity of SOD in group A, C and D were remarkablely higher than the group2K1C.③The content of MDA and SOD in cochlear tissues of group A,C and D were decreased markedly(P<0. 05)than group 2K1C. ④Sodium salicylate reduced the incidence rates of DPOAE in low level stimulus intensity significantly expect shame group in rats with renovascular hypertension.⑤The amplitudes of DPOAE in group B and C were undiffer entiated with groupA(P>0.05) ;but in group C was decreased significantly than group A at every frequency(P<0. 01). ⑥The latencies of DPOAEwere not affected by sodium salicylate statistically in four groups. Conclusion ①The effects of sodium salicylate on the distortion product otoacoustic emission( DPOAE) in rats with renovascular hypertension by change cochlear microcirculation. ②Sodium salicylate can effect metabolism of free radical in rats with renovascular hypertension, but it is not related to the effects of sodium salicylate on the DPOAE.
