艾氏腹水瘤细胞热休克蛋白70的抗瘤效应

摘要 目的 观察从艾氏腹水瘤细胞胞浆中提纯的热休克蛋白 70 (heatshockprotein70 ,HSP70 )的抑瘤作用 ,并探讨其机制。方法 用亲和层析提纯艾氏腹水瘤细胞的HSP70 ,应用体内免疫法检测HSP70的抑瘤作用 ,用3 H -TdR释放法检测特异性细胞毒T淋巴细胞 (CTL)的杀伤活性。结果 与对照组相比免疫组小鼠平均肿瘤重量明显减轻、体积明显缩小 ,特异性CTL的杀伤活性显著增强。结论 HSP70可诱导机体产生特异性杀伤活性 。 Objective In order to investigate the tumor-specific cytotoxicity induced by HSP70 peptide derived from Ehrlich ascites tumor cell and to study its mechanisms. Methods An animal model of tumor vaccination was established and the average weight and the diameter of the tumor no dules of tumor-bearing BALB/c mice after immunization were observed.The 3H-TdR release assay was used to examine the cytotoxicities of the splenocytes to Ehrlich ascites tumor cells. Results It showed that specific cytotoxicity of splenocytes from the immunized mice to the Ehrlich ascites tumor cells was significantly higher than that from the tumor-bearing and control groups . Conclusions HSP70 Ehrlich ascites tumor cell are active in inhibiting tumor and could be used in effective anti-tumor therapy.
