摘要
把pMTSA,pHBV3.0,pSMGHI和pMDV38kdpp4种重组质粒,以单个或二者混合后经微注射导入家畜的受精卵,随后移入同种寄母,获得转基因家畜,兔导入上述外源基因的卵,移植后妊娠率59.70%(40/67),发育成个体率17.74%(129/727),整合率59.38%(57/96),在整合兔中有32.14%(9/28)表达;山羊,导入pSMGHI基因卵,移植后妊娠率50%(7/14)发育成个体率12.76%(12/94),整合率33.33%(4/12);牛,获一整合的流产胎儿。当代转基因兔的后代也检出整合的外源基因,并有部分表达,证明导入的外源基因是可以遗传的。研究表明,这一转基因系统具有相当的稳定性,并可应用家畜代替常规的生物反应器,生产具较高经济价值的蛋白质,为畜牧生产开辟了一条新途径。
Livestock Zygotes microinjected with one or two gene constructs containing the pMTSA, pHBV 3.0, pSMGHI and pMDV38kdpp gene were transferred to the same species host mother. Transgenic rabbits, goats, and cattle have been produced. After rabbit zygotes microinjected above foreign genes were transferred, the percentage of pregancy of rabbits is 59.70% (40/67), the ratio of the embryos to develop into fetuses is 17.74% (129/727), the ratio of integration is 59.38% (57/96), and the expressive ratio is 32.10% (9/28), goat zygotes microinjected with pSMGHI gene were transferred, the pregnant percentage is 50% (7/14), the embryos developed to fetuses is 12.76% (12/94), the ratio of integration is 33.33% (4/12). A transgenic cattle aborted fetus was obtained by microinjecting pMTSA gene into cattle zygotes. The first and second generations of transgenic rabbits have been integrated the foreign gene. Some of them expressed, too, so the foreign genes have transmitted into progenies. Evidences showed above indicated that this transegenic system is reliable and the conventional bioreactor can be replaced by livestock to produce a vast economic protcins. A new way have been opened up for the animal production.
