Establishing Embryogenic Cell Suspension Cultures in Wheat by An Alternative Simple Method

摘要 源自普通小麦中国春幼胚的胚性愈伤组织经4—5个月继代培养后仍保持胚性。选取没有明显形态分化的致密胚性愈伤组织进行悬浮培养;通过向培养基中添加活性炭(AC)和延长继代间隔至一个月的途径,成功建立分散性好、生长速度快、稳定的胚性悬浮细胞系。对其生长特性观测表明,旺盛生长期发生于继代后4—12天;PCV加倍时间为5天。初步实验表明,该细胞系具有形态发生能力,并可游离出大量有活力原生质体,可作为体细胞遗传学研究的良好材料。 The embryogenic callus of Triticum aestivum L. (cv. Chinese Spring) which had been subcultured for 4 to 5 months was selected as the source material for liquid culture. By involving activated charcoal in the liquid MS medium and extending subculture intervals to one month, finely dispersed, fast growing and stable embryogenic cell suspension cultures were established. The growth characteristics of the cell suspension cultures were studied.
