摘要
为寻找到快捷有效的生理生化抗药性检测方法,对供试的6株对速克灵具有不同抗性水平的核盘菌进行了可溶性蛋白和酯酶的聚丙烯酰胺凝胶电泳谱带的试验研究。结果表明:不同抗性水平的菌株间蛋白质及酯酶图谱存在明显差异,中抗菌株与敏感菌株可溶性蛋白的主带基本一致,但中抗菌株的带数略多于敏感菌株,Rf=0.14,Rf=0.53两条带为中抗菌株所拥有,而敏感菌株不具有,高抗菌株的谱带数又显著少于敏感菌株,Rf=0.53的谱带为抗性菌株所特有。酯酶电泳图谱中敏感菌株与抗性菌株均存在特异性酶带,敏感菌株在Rf=0.20处有一条主酶带是抗性菌株所没有的,Rf=0.22的主酶带则为抗性菌株中所特有而敏感菌株没有的。因此,认为Rf=0.20的酯酶带与核盘菌的抗药性有一定相关性。
In order to find an effective physiological and biochemical method to detect fungicide resistance, electro-phoretic bands and patterns of the soluble proteins and esterases from six isolates of Sclerotinia sclerotiorum with different resistant levels to procymidone were determinded with the method of poyacylamide slab gel electrophoresis. The results showed that the electrophoretic patterns of proteins and esterases from various strains were significantly different. The soluble proteins from moderate resistant isolates had the similar main bands with sensitive isolates, but had more bands than sensitivity ones, which (Rf=0. 14, Rf =0. 53) existed in moderate isolates. The high-level resistant strains stably had fewer bands than sensitive ones. The band (Rf=0. 53) was only found in resistant isolates. Resistant and sensitive isolates had their particular esterase bands. Sensitive isolate had a main band (Rf=0. 20), which did not exist in resistant isolate, while the band (Rf =0. 22) could only be found in resistant isolate. Therefore, the band (Rf =0. 22) was considered to be correlative to the resistance of Sclerotinia sclerotiorum.
