摘要
以12个枇杷栽培品种为试材,用光学显微镜和电子显微镜观测新鲜花粉、戊二醛固定花粉、风干花粉和醋酸酐分解花粉的形态及大小,比较不同制样处理花粉的扫描效果,结果表明,1戊二醛固定花粉、醋酸酐分解花粉与新鲜花粉的形状、大小均无差异,花粉赤道面观均为近长球型或长球形,极面观近圆形或三角形,表面纹饰均清晰可见。2风干花粉与戊二醛固定花粉相比,极轴平均增长15.49%,赤道轴平均缩短18.85%,P/E比值平均增大43.09%。风干花粉赤道面观7个品种为超长球形,其余为长球形;3与其他制样方法相比,戊二醛固定的处理,操作简便,成本低,观察时间不受季节等影响,扫描效果及花粉的形状、大小与新鲜花粉均无差异,是进行枇杷孢粉学研究较理想的方法。
The shape and size of fresh, air\|dried(AD), glutaric aldehyde fixed(GAF) and acetic oxide decomposed(AOD) pollens of 12 loquat varieties were compared in the test.The results showed that,there were no differences in shape and size among fresh, GAF and AOD pollens.The polar axiss of AD pollen increased by 1549 percent and the Equatorial diameter decreased by 1885 percent in comparison with GAF pollen.The shape of AD pollen of 7 varieties changed into perprolate but GAF pollen was all prolate.The method of GAF had good scanning result and low cost, it was easier in operation than the method of AOD, and could be the ideal way in the palynological study of loquat.
