姜黄素联合化疗药对HepG2细胞PI3K信号通路的影响

Effect of PI3K / AKT signaling pathway and related regulatory molecular of HepG2 cell intervened by curcumin combining with 5-fluorouracil

摘要 目的研究5-氟尿嘧啶(5-FU)单用或联合姜黄素对Hep G2细胞PI3K/AKT信号通路及相关调控分子AKT、PTEN和TLR4的影响。方法 5-FU单用或联合姜黄素对Hep G2细胞PI3K/AKT信号通路及相关调控分子AKT、PTEN和TLR4的影响。结果姜黄素可以促进Hep G2细胞的凋亡,随着浓度的增加和时间的延长促凋亡作用逐渐明显。联合5-FU的促凋亡作用更加明显。姜黄素联合5-FU通过PI3K/AKT信号通路降低了AKT的水平而提高了PTEN、TLR4的水平,从而提高了抗肿瘤的作用。结论姜黄素能够明显加强肝癌细胞对5-氟尿嘧啶的药物敏感性,抑制了肝癌细胞的生长,促进了肿瘤细胞的凋亡,提高了抗肿瘤的作用。 Objective To study the effect of 5-FU alone or combined with curcumin on HepG2 cells of PI3K/AKT signal pathway and related regulatory.Methods The experiment was divided into two parts.The first part:the effets of 5-FU a-lone or combined with curcumin on HepG2 cells of PI3K/AKT signal pathway and related regulatory molecules AKT,PTEN and TLR4.Results Curcumin can induce apoptosis of HepG2 cells,with concentration and time increasing apoptosis gradually clear.Apoptosis of 5-FU is more obvious.The effect of curcumin and 5-FU through PI3K/AKT signaling reduced AKT levels and increased PTEN,TLR4 level,thereby improving the antitumor effect.Conclusion Curcumin can enhance the susceptibili-ty of liver cancer cells on 5-fluorouracil,inhibited the growth of liver cancer cells,promote the apoptosis of tumor cells,improve the anti-tumor effect.
