抑癌基因PTEN及胚胎性转录因子FOXC2在晚期上皮性卵巢癌组织中的表达及与临床病理特征的关系

摘要 目的分析抑癌基因PTEN及胚胎性转录因子FOXC2在晚期上皮性卵巢癌组织中的表达及与临床病理特征的关系。方法选取2016年2月-2018年2月在医院接受手术治疗的晚期上皮性卵巢癌患者50例。分别采集卵巢癌组织及癌旁正常组织标本,采用免疫组化法检测不同组织中PTEN与FOXC2的表达,分析PTEN和FOXC2与上皮性卵巢癌患者临床病理特征的关系。结果上皮性卵巢癌组织中PTEN的阳性表达率为52.00%明显低于癌旁正常组织的100.00%,而FOXC2的阳性表达率为88.00%明显高于癌旁正常组织的16.00%,差异均有统计学意义(P<0.05)。分化程度为中高分化、临床分期为Ⅰ~Ⅱ期、无淋巴结转移的上皮性卵巢癌组织中PTEN阳性表达率明显高于分化程度为低分化、临床分期为Ⅲ~Ⅳ期、有淋巴结转移者(P<0.05)。临床分期为Ⅰ~Ⅱ期、无淋巴结转移的上皮性卵巢癌组织中FOXC2阳性表达率明显低于临床分期为Ⅲ~Ⅳ期、有淋巴结转移者(P<0.05)。结论抑癌基因PTEN在上皮性卵巢癌组织中呈明显低表达,胚胎性转录因子FOXC2在上皮性卵巢癌组织中呈明显高表达。且PTEN、FOXC2与上皮性卵巢癌的发生、发展、分化程度及淋巴结转移存在密切相关,可能成为临床上诊断上皮性卵巢癌的敏感指标。 Objective To analyze the expression of tumor suppressor gene PTEN and embryonic transcription factor FOXC2 in advanced epithelial ovarian cancer and their relationship with clinical and pathological features.Methods 50 patients with epithelial ovarian cancer who received surgical treatment in our hospital from February 2016 to February 2018 were selected.Ovarian cancer tissues and normal adjacent tissues were collected as specimens,and the expression of PTEN and FOXC2 in different tissues was detected by immunohistochemistry,and the relationship between the above two indicators and clinical features of epithelial ovarian cancer patients was analyzed.Results The positive expression rate of PTEN in epithelial ovarian cancer tissues(52.00%)was significantly lower than that in adjacent normal tissues(100.0%),the positive expression rate of FOXC2(88.00%)was significantly higher than of normal tissues adjacent to the cancer(16.00%),the differences were statistically significant(P<0.05).Differentiation degree of high differentiation,clinical stage wasⅠ~Ⅱ,without lymph node metastasis of epithelial ovarian cancer tissue of PTEN positive rate was higher than low differentiated tissue differentiation,clinical stage wasⅢ~Ⅳ,with lymph node metastasis was higher(P<0.05).Clinical stage wasⅠ~Ⅱ,without lymph node metastasis of epithelial ovarian cancer tissues FOXC2 positive rate compared with the clinical stage forⅢ~Ⅳ,lymph node metastasis was lower(P<0.05).Conclusion The tumor suppressor gene PTEN is significantly low expressed in epithelial ovarian cancer,and the embryonic transcription factor FOXC2 is highly expressed in epithelial ovarian cancer.In addition,PTEN and FOXC2 are closely related to the occurrence,development,differentiation and lymph node metastasis of epithelial ovarian cancer,which may be a sensitive indicator for clinical diagnosis of epithelial ovarian cancer.
