摘要
杀鲑气单胞菌(Aeromonas salmonicida)是鲑类疑难细菌性肾脏病原菌之一。用该菌免疫BALB/c鼠,取其脾细胞与SP_2/O骨髓瘤细胞融合。酶联免疫吸附分析法(ELISA)及萤光抗体法(FAT)筛选所需单克隆抗体。有限稀释法克隆化建立了一株抗杀鲑气单胞菌抗原的单克隆抗体杂交瘤细胞株。免疫球蛋白亚类鉴定为lgG_1,该单克隆抗体(McAb)与16份确诊为杀鲑气单胞菌肾脏病热提取抗原反应,具有高度特异性识别抗原作用。
Aeromonas Salomonicida is one of the bacterial kidney diseases of salmonids which is difficult to identify. The mice of BALB/C were immunized with this bacterium. The speen cells of the mice were fused with myeloma cells of SP_2/O. Needed monoclonal antibody was screened by Enzyme-linked Immunosorbent Assay(ELISA) and Fluorecent Antibody Technic (FAT). After two cloning by limiting dilution, the cell of myeloma which produced monoclonal antibody against the antigen of A. salmonicida was gotten. The McAb is IgGl. The McAb has high speciality to identify the antigen of A. salmonicid a which was known bacterium of kidney disease in salmonids.
