姜黄素、阿米洛利联用对大鼠肝星状细胞增殖的影响

Effect of the Combination Use of Curcumin and Amiloride on the Proliferation of Rat Hepatic Stellate Cells

摘要 目的 探讨姜黄素 (curcumin)、阿米洛利 (Amiloride)联用对大鼠肝星状细胞增殖的影响。方法 HSC T6细胞与不同浓度的姜黄素、阿米洛利共同培养 2 4h。LDH法观察姜黄素、阿米洛利对HSC T6的毒性作用 ,以MTT法观察姜黄素、阿米洛利单用及联用对HSC T6增殖的效应。结果 各治疗组HSC的增殖与对照组相比明显为低 ,均有显著性差异 (P <0 0 0 1) ;联用组对HSC增殖的抑制率最高 ;联用组HSC的增殖更低于姜黄素组 (P <0 0 5 )。各治疗组培养上清液中LDH活力与对照组相比未见显著性差异 (P >0 0 5 )。结论 姜黄素、阿米洛利均可抑制HSC的增殖 ,且联用组优于姜黄素单用组 ;它们的作用不是非特异性细胞毒作用所致。 Objective To investigate the effect of the combination use of curcumin and amiloride (CUCA) on the proliferation of rat hepatic stellate cells (HSC). Methods HSC T6 were cultured in different concentrations of curcumin and amiloride for 24 hours. LDH colorimetry was used to observe the cytotoxicity of curcumin and amiloride on HSC T6. MTT colorimetry was used to observe the effects of the respective use and the combination use of curcumin and amiloride on the proliferation of HSC. Results It was found that the HSC proliferation rate in each drug treating group was markedly lower than that in the control group (P<0.001); the proliferation inhibitory rate in the group treated by CUCA was the highest. It was also found that the HSC proliferation rate in the CUCA treating group was much lower than that in the curcumin treating group ( P <0 05). The LDH activity detected in the supernate of each drug treating group showed no marked difference as compared with that in the control group ( P >0 05). Conclusion Both curcumin and amiloride can inhibit the HSC proliferation, and the inhibitory effect of CUCA is better than that of curcumin alone; the effects of the two drugs are not of nonspecific cytotoxicity.
