向日葵总DNA不同提取方法比较及在RAPD中的应用研究 被引量:6

摘要 比较了CTAB法和SDS法从油用向日葵的真叶和种子中提取DNA的提取过程、时间、DNA质量及在RAPD PCR中应用的效果 ,结果表明 :不同方法及材料中均能获得质量较好的DNA ,而且在RAPD PCR中均获得了较好的扩增结果。但利用SDS法从种子中提取DNA简捷、快速 ,且不受取材的限制 ,可满足RAPD技术处理大量样品的要求 。 In this paper, results of DNA extracted from the first or second young leaf and seed of oil sunflower with CTAB and SDS methods were compared in their procedures, time consumed, DNA quality and quantity, and application in RAPD PCR.The results showed that total DNA with high quality could be obtained both from different methods and different materials. Meantime, ideal amplification bands could be got in RAPD PCR reaction with all these DNA. But among these results, we considered that SDS method using seed as material is much simpler and quicker in procedure, and there is no problem with sampling time, so it can meet the needs for handling large quantity of samples in RAPD PCR research.
