摘要
利用农杆菌介导方法〔9〕,将ACC脱氨酶基因〔1 0 ,1 2〕导入新疆哈密瓜的两个品种 :皇后和卡拉克塞 (伽师瓜 )。以 5d的无菌子叶作为外植体 ,在浓度OD =0 .4的菌液中侵染 5~ 8min ,共培养 2d ,经过脱菌、分化、继代及生根培养后 ,得到完整的抗卡那霉素转化植株。移栽试验表明 ,炼苗后经过水培的植株其成活率高 ,且后期生长较迅速。
A gene which encodes 1-aminocyclopropane-1-carboxylic aci d deaminase (ACCd) was transfered into Hami melon(Cucumis melo.L) of Xinjiang (Huanghou and Jiashigua) by Agrobacterium tumefaciens-mediated method was studied.The kanamycin-resistant transgenic plants that were regenerated fro m cotyledon explants of 5-day-old were infected 5~8 min in Agrobacterium tume faciens, co-cultured 2 days, and then differentiation and root culture by using debacterium.The frequency of bud differentiation can be increased with AS and AgNO 3 in culture medium. The experiments of transplanting showed that the surv ival rate of plants by trained and water-cultured method was higher and the gro wth of later stage was faster. The rooting and growing rates of transgenic plant s were less than that of controlled plants.
