应用浸种法导入外源DNA转化烤烟遗传性状变异D_2代研究(Ⅰ)

摘要 提取抗赤星病烤烟 CV 87的总 DNA,采用浸种法导入感赤星病烤烟 NC89。 D_1 代筛选出的优良变异株收种并种植后得到 D_2代 ,对其活体接种赤星病菌 ,检测其抗病性 ,并于接菌前后测定叶片过氧化物酶活性。对其田间生长性状调查 ,对其成熟期烟叶总糖、蛋白质及烟碱分析。结果表明 ,浸种法直接导入外源 DNA ,在变异株 D_2代仍可有效转移株高、株型、叶型、叶面积及抗病性等性状 ,并获得较高的转化率 ,D_2代为 8.10 %。高抗赤星病 ,过氧化物酶活性高 ,总糖、蛋白质及烟碱含量基本保持了受体原有的优良品质 ,其变异株 D_2代有 C_10,A_9,B_6,D_3,A_13。 Researched on the D 2 generation gained from the good strains of D 1 generation that translated the total DNA of Red Star Disease resistance tobacco CV87 into Red Star Disease infection NC89 through seed immersion method and analyzed its POD activity,total sugar, protein,nicotine,the result showed that:the phenotypes of D 2 generation such as height,type,leave area,growth period and disease resistance,etc,were effectively transferred into D 2 generation and transform rate was 8.10%.Having higher disease resistance,higher POD activity,and keeping the total sugar,protein,nicotine level with receptor's good character in the D 2 generation,the mutant D 2 generation were C 10 ,A 9,B 6,D 3,A 13 ?$$$$
