抗凝血酶Ⅲ的稳定性考察

摘要 目的研究温度和光照对抗凝血酶Ⅲ(AT-Ⅲ)生物活性的影响。方法设置不同温度(25±1)、(40±1)、(60±1)、(70±1)℃和(4500±500)l X的强光直接照射,样品放置不同时间,用凝血酶空斑法测定样品的AT-Ⅲ活性。结果 AT-Ⅲ具有一定的耐热性,在25℃左右可放置3个月活性无明显变化,随着时间的延长活性有下降的趋势。40℃放置12d活性变化很小,48d活性损失30.6%;60℃放置12 h活性损失8.8%;70℃以上放置时AT-Ⅲ活性迅速丧失,10 min活性降低约33%,15 min活性下降约50%。此外,AT-Ⅲ对直接光照较敏感,光照10 d活性明显下降。结论温度和光照对AT-Ⅲ活性有明显影响,因此该制品最好在4~8℃避光处放置。 Objective To investigate the effect of temperature and light on biological activity of antithrombin Ⅲ(AT-Ⅲ), in order to provide evidence for the stability of AT-Ⅲ. Methods The activity of AT-Ⅲ was detected by thrombin gel lacuna and different times for depositing samples under different temperatures(25±1)℃、(40±1)℃、(60±1)℃、(70±1)℃, as well as samples exposed to light at(4500 ±500)lx. Results AT-Ⅲ was provided with difinite heat-resistant. The Activity of AT-Ⅲ didn't change significantly at(25±1)℃ for 3 months, but the activity was decreased with prolongation of time. The activity of AT-Ⅲ changed a little at 40℃ for 12days,and the activity lost 31.6% for 48 days. The activity decreased 10% at 60℃ for 12h. AT-Ⅲ rapidly lost its avtivity at 70℃, the activity reduced to 55.8% for 15min. Conclusion The activity of AT-Ⅲ is significantly influenced by temperature and light, so the best storege temperature of AT-Ⅲ preparation is 4~8℃.
