苯酚-次氯酸盐法测定空白脂质体透析液中铵离子浓度 被引量:3

摘要 目的建立空白脂质体透析液中硫酸铵的定量测定方法。方法将样品依次加入显色剂I、显色剂II显色后,在波长630 nm处比色,并进行了线性、精密度、稳定性和加样回收率等项考察。结果该方法线性方程为A=72 035ρ+4.4,质量浓度在1~20 mg·L–1内,线性关系良好(r=0.999 7),该方法精密度为2.65%;显色后2 h比色结果稳定;加样回收率为97.3%~99.5%。结论该方法适于空白脂质体透析液中硫酸铵含量的测定。 Objective To develop a method that can be used to determine the concentration of ammonium ion in external environment of blank liposome. Methods Added chromogenic agent I, II into the sample, and then bathing at 37 ℃ for 30 min,at last colorimetered at 630 nm. In addition, the precision, stability, and the recovery of the method have been checked. Results The equation was A=72 035 ρ + 4.4,with a good correlation in the concentration range of 1-20 mg·L-1(r = 0.999 7). The precision of the method was 2.65%(RSD), the absorption value was stable in 2 h after coloration. Conclusion This method is simple, creditability and was feasible to determine the concentration of ammonium ion in the external environment of blank liposome with good precision and reproducibility.
