摘要 目的制备克里唑替尼(crizotinib,CIB)脂质体,并建立CIB的含量测定方法。方法采用p H梯度法制备克里唑替尼脂质体(crizotinib liposomes,CIBL),以紫外分光光度法测定脂质体中CIB的含量。结果实验条件下所用辅料对CIB的测定无干扰,CIB对照溶液的质量浓度在15.0~35.0 mg·L^(-1)内线性关系良好,相关系数r=0.999 6,回收率在99.8%~101.2%内,日内及日间精密度RSD均小于2%(n=3),制得CIBL的平均包封率为90.8%±1.11%。结论 pH梯度法可用于制备CIBL;紫外分光光度法测定CIB的含量简便易行,结果准确,可用于CIBL的含量测定。 Objective To prepare crizotinib(CIB) liposomes and establish a method for determination of its content. Methods Crizotinib liposomes were prepared by p H gradient method. Ultraviolet spectrophotometry was used to determine its content with the detection wavelength of 336 nm. Results Adjuvants used under the experiment conditions did not interfere with the determination results of CIB. CIB had a good linear relationship over the range of 15.0-35.0 mg·L^(-1)(r=0.999 6), the recovery was in a range of 99.8%-101.2%, the intra-day RSD and inter-day RSD were less than 2%(n=3). The average entrapment efficiency of CIB liposomes were 90.8% ± 1.11%. Conclusions p H gradient method could be used to prepare CIB liposomes. It is a simple and accurate method to determine the content of CIB in the liposmes by ultraviolet spectrophotometry.
