溶血栓新药瑞替普酶生化特性确证的研究 被引量:2

摘要 对溶血栓新药瑞替普酶(reteplase)的鉴别和纯度测定是该基因工程药物的科研试验与工业化生产中的重要内容。采用薄层扫描、S2251发色底物法、SDS PAGE银染、ProteinC4RP HPLC分析柱检测、N端15个氨基酸残基分析以及激光解析飞行时间质谱分析等实验,证明笔者克隆的瑞替普酶与标准品的生化特性一致,并对工业生产中的常规检测提出了建议。 It is very importmant to confirm biochemical properties of Reteplase in lab and factories.The biochemical properties of reteplase were determined using some methods,i.e.molecular weight was determined using matrix assistance laser desorption ionization massspectrometry (MALIDMS),purity was analyized by silver staining SDSPage and protein C4 RPHPLC,amino acid sequence was analyzed using the amino acid sequence analyzer of an ABIPrcise491,reteplase activity was measured using the spectrophotometric assay method,etc.Some of them were suggest as normal methods in factories.The results were confirmed that our reteplase is the same as to the control of Retevase.
