摘要
目的 复制大鼠慢性应激性抑郁症模型 ,观察大鼠血清对淋巴细胞转化的抑制作用及其与抑郁症的相互关系。方法 以 8种不可预料的中等强度应激作用于大鼠 ,每天 1次共 16d ,制备大鼠抑郁症模型 ,采用开场试验检测大鼠的行为性抑郁 ,用MTT法测定淋巴细胞转化。结果 大鼠接受慢性应激的第 14天出现显著的行为性抑郁 ,在停止应激后的第 12天 ,行为性抑郁仍维持较高程度 ;而抑郁症大鼠血清对刀豆素 (ConA)诱导的淋巴细胞转化的抑制作用 ,也是从应激后的第 14天开始 ,在停止应激后的第 12天抑制率仍为 2 5 %。结论 慢性应激引起大鼠的行为性抑郁 ,停止应激后 ,至少在 2周内存在抑郁性行为。应激性抑郁症与大鼠血清对正常淋巴细胞转化的抑制作用相平行。
Objective To study the relationship between depressive severity and the effect of serum on lymphocyte proliferation in depressive rats induced by chronic stresses. Methods Depression animal model was reproduced with 8 unexpectable stressors in 16 days and the depressive behavior was measured with Open Field test. Lymphocyte proliferation was tested with MTT method. Results The depressive behavior started from day 14th after stress, reached the peak at day 21th and at least lasted to day 28th. The serum from depressive rat inhibited ConA-induced lymphocyte proliferation which paralleled to depressive severity. Conclusion Chronic stress could induce depressive behavior in rats, and which is related with the inhibitory effect of serum on lymphocyte proliferation.
