苦参碱触发的K562细胞胞内钙信号的动态变化 被引量:8

摘要 目的 研究苦参碱诱导K5 6 2细胞分化的信号转导机制。方法 以Fura 2 AM为荧光探针 ,定量分析苦参碱作用后K5 6 2细胞内钙离子浓度的动态变化。结果 经苦参碱处理后 ,在胞外有钙的情况下 ,K5 6 2细胞内钙离子浓度由 12 9.0 2nmol/L升至 2 0 7.5 0nmol/L ,并维持较长时间 ;在无胞外钙的情况下 ,胞内钙离子浓度由 12 9.0 2nmol/L迅速升至 16 4 .2 2nmol/L ,并迅速恢复。两者的增高幅度均与苦参碱浓度呈正相关。结论 在苦参碱诱导K5 6 2细胞分化过程中 。 Objective To study the mechanism of signal transduction in K562 cells differentiation induced by matrine.Methods Fura 2 AM, a sort of fluorescent probe for free calcium, was used to analysis the dynamic change of the calcium signaling in K562 cells treated by matrine.Results After K562 cells were treated by matrine,concentration of the free calcium in K562 cells without extracellular calcium increased from 129.02 nmol/L to 207.50 nmol/L and kept the high level for a relative long time, while it increased rapidly from 129.02 nmol/L to 164.22 nmol/L 0and had a quick comeback. Both of the increased extents are in response to concentration of the matrine.Conclusion It is an immediately important signaling that the change of concentration of free calcium involved in proliferation suppression and cell differentiation of K562 cells induced by matrine.
