双歧杆菌原生质体制备条件的探讨 被引量:2

摘要 目的 探讨Bifidobacterium(双歧杆菌 )原生质体制备的条件。方法 将培养至对数生长期的双歧杆菌用α 淀粉酶和溶菌酶进行酶解脱壁。结果 在 37℃条件下 ,80 0 μg/ml溶菌酶和 5 0 0 μg/mlα 淀粉酶用于双歧杆菌联合酶解脱壁 6 0min ,双歧杆菌原生质体形成率为 80 %左右。结论 α Objective To Study on Protoplast formation in bifidobacterium .Method Cell walls of bifidobactera were detached from cell membranes by hydrolyzation with alpha amylase and Lysozyme together when they were fermented to the period of logarithm growth.Result Protoplast formation frequency was 80% under the condition that bifidobactera were incubated with 500 μg/ml alpha amylase and 800 μg/ml Lysozyme at 37℃ in 60 min.Conclusion Alpha amylase in combination with Lysozyme is facilitate to Protoplast formation of bifidobacterium .
