睫状神经营养因子对N-甲基-D-天冬氨酸引起海马神经元内钙离子浓度升高的影响 被引量:1

摘要 ①目的 研究睫状神经营养因子 (CNTF)对谷氨酸 (Glu)受体激动剂N 甲基 D 天冬氨酸 (NMDA)引起的海马神经元内游离Ca2 + 浓度 ([Ca2 + ]i)升高的影响 ,并探讨G蛋白是否参与此作用。②方法 原代培养海马神经元 ,用活细胞内荧光探针Fura 2 /AM实时检测应用CNTF和G蛋白抑制剂百日咳毒素 (PTX)后由NMDA引起的海马神经元 [Ca2 + ]i 的变化。③结果 用CNTF孵育经NMDA刺激的海马神经元后 ,细胞内 [Ca2 + ]i 升高的程度较单独NMDA刺激细胞引起的 [Ca2 + ]i 升高程度低 (t=2 .97~ 4 .86 ,P <0 .0 5 ) ,加入PTX可减弱CNTF的抑制作用。④结论 G蛋白可能参与CNTF调节NMDA引起的海马神经元内 [Ca2 + ]i Objective\ To investigate the effect of ciliary neurotrophic factor (CNTF) on the increase of [Ca 2+ ] i in hippocampus induced by N methylaspartate (NMDA) and observe whether GTP binding proteins participate in this pathway.\ Methods The primary hippocampal neurons were cultured and [Ca 2+ ] i in hippocampal neurons treated with NMDA and incubated with or without CNTF as well as pertussis toxin(PTX) were measured with the calcium sensitive fluorescent dye Fura 2/AM.\ Results The increase of [Ca 2+ ] i induced by NMDA after the hippocampal neurons were incubated with CNTF was lower than that with NMDA alone( t=2.97-4.86,P <0.05),PTX could reduce the inhibitory effect of CNTF.\ Conclusion\ CNTF could inhibit the increase of [Ca 2+ ] i induced by NMDA, this rapid action of CNTF is related to the GTP binding proteins.
