摘要
目的 :探讨 P4 4 / 4 2 MAPK在 γ射线诱发的白血病小鼠骨髓和胸腺细胞中的表达及其作用。 方法 :建立 6 0 Coγ射线体外诱发 BAL B/ c小鼠白血病模型 ;分离白血病组辐射未癌变组和对照组小鼠胸腺和骨髓细胞总蛋白 ,应用蛋白印迹法分析各组细胞 P4 4 / 4 2 MAPK的表达及磷酸化水平。 结果 :白血病小鼠骨髓和胸腺细胞 P4 4 / 4 2 MAPK的蛋白表达均显著高于未癌变组和对照组 (P<0 .0 5 ) ;在 3组小鼠中 ,白血病小鼠的胸腺和骨髓细胞 P4 4 / 4 2 MAPK磷酸化水平最高 (P<0 .0 5 )。结论 :提示 P4 4 / 4 2
Objective: To investigate the role of the P44/42 mitogen activated protein kinases in leukemia cells induced by γ ray irradiation. Methods:Mouse leukemia model was induced by 60 Co γ ray irradiation: The mice were divided into 3 groups: the leukemia group, the uncancerization group and the unirradiated control group according to the pathological examination. Western blotting was performed to detect the P44/42MAPK expression level and the phosphoryation level in the mice thymus and bone marrow cells. Results: The P44/42MAPK expression was significantly higher in the thymus and marrow tissues of the leukemia group than that of the uncancerization and control groups( P <0.05).The level of P44/42MAPK phosphorylation in leukemia group was higher than that of the uncancerization and control groups( P <0.05). Conclusion: P44/42MAPK may play a role in proliferation disorder of leukemia cells induced by γ ray irradiation.
