摘要
目的 :研究作为氧化应激蛋白的血红素加氧酶 (hem e oxygenase,HO)对氧惊厥发生的影响。方法 :雄性 SD大鼠 4 0只 ,随机分为正常对照组、氧惊厥组、Na2 CO3处理组、锌卟啉 (Zn- PP)处理 4 h组和 Zn- PP处理 16 h组 ,观察应用 HO特异性抑制剂 Zn- PP后氧惊厥始发时间的变化 ,同时测定 HO活性以及反映机体氧化损伤的指标丙二醛 (MDA)的含量。结果 :氧惊厥组 HO活性为 (3.4 36± 0 .4 90 )μm ol/ (g· h) ,比对照组显著增强 (P<0 .0 1) ,Zn- PP处理 4 h组和 Zn- PP处理 16 h组 HO活性分别为 (1.16 9± 0 .12 2 )、(1.5 91± 0 .191)μm ol/ (g· h) ,较氧惊厥组显著降低 (P<0 .0 1)。Zn- PP处理 4 h组 ,Zn- PP处理 16h组的惊厥始发时间分别为 (4 0 .4 7± 4 .6 2 )、(30 .15± 4 .5 9) min,较氧惊厥组显著延长 (P<0 .0 1)。 Zn- PP处理 4 h组 MDA量较其他各组显著增高 (P<0 .0 1)。 结论 :氧化应激引起的
Objective: To investigate whether heme oxygenase, the major anti oxdative protein, has effect on the onset of hyperoxia induced seizures. Methods: Rats were divided into control group, convulsion group, Na 2CO 3 treated group, Zn PP treated 4 h group and 16 h group. The changes of the initial time of convulsion were observed after using zinc protoporphyrin Ⅸ, a specific inhibitor of heme oxygenase. At the same time, the activity of heme oxygenase was measured and the concentrations of methylenedioxyamphetamine, a parameter of oxdative injury, were also measured. Results: The activity of heme oxygenase in convulsion group was significantly increased[(3.436±0.490) μmol/(g·h)] compared with that of control group( P <0.01); while the activity of Zn PP treated 4 h group and 16 h group were (1.169±0.122) and (1.591±0.191) μmol/(g·h) respectively, which were significantly decreased compared with that of control group( P <0.01). The initial time of convulsion in Zn PP treated 4 h group and 16 h group were (40.47±4.62) and (30.15±4.59) min respectively, which were significantly prolonged compared with convulsion group( P <0.01).The concentration of methylenedioxyamphetamine in Zn PP treated 4 h group was significantly increased ( P <0.01). Conclusion: The increase of heme oxygenase activity caused by oxdative stress may promote the onset of hyperoxia induced seizures.
