摘要
目的 :探讨γ射线体外诱发的白血病小鼠胸腺细胞总蛋白质酪氨酸磷酸化水平的变化。方法 :35 3只 BAL B/ c小鼠分为照射组及对照组 ,照射组经 6 0 Coγ射线全身照射后 ,病理证实出现白血病者为癌变组 ,未出现任何肿瘤者为未癌变组 ;对照组为正常 BAL B/ c小鼠 ,未经γ射线照射 ,与照射组同步喂养。分离胸腺细胞 ,应用免疫沉淀法及 Western印迹分析检测蛋白质酪氨酸磷酸化水平。 结果 :癌变组胸腺细胞中蛋白质酪氨酸磷酸化水平总体上明显高于对照组及未癌变组 ,尤以相对分子质量 (4 7.5~ 83)× 10 3及 32 .5× 10 3附近蛋白变化最为明显 ;相对分子质量为 35× 10 3的蛋白在癌变组中酪氨酸磷酸化明显 ,而在对照组及未癌变组中未检测到磷酸化的发生。 结论
Objective:To investigate change of tyrosine phosphorylation in γ ray irradiation induced thymus leukemia in mice. Methods:A total of 353 BALB/c mice were divided into irradiation groups and control group. Irradiation groups, which were radiated with γ ray irradiation, included cancerization group confirmed with histology and uncancerization group. Control group was fed synchronistically with irradiation groups. Tyrosine phosphorylation was detected with Western blot and immunoprecipitation in thymus cells. Results: Tyrosine phosphorylation in cancerization group was significantly higher than that in uncancerization group and control group, especially in the protein with molecular weight of (47.5 83)×10 3 and close to 32.5×10 3. And tyrosine phosphorylation of molecular weight of 35×10 3 was very high in canerization group, but it was not found in uncancerization group and control group. The results detected with Western blot and immunoprecipitation were similar. Conclusion: Tyrosine phosphorylation is significantly increased in cancerization group, specially in the protein with molecular weight of (47.5 83)×10 3 and close to 32.5×10 3, suggesting that they may be correlated with γ ray irradiation induced leukemia in mice.
