摘要
以新鲜幼嫩的新疆雪莲无菌叶片作外植体,接种于含有不同激素水平的MS培养基上,经15d后,愈伤组织形成,并分化出很多丛生芽.在诱导愈伤组织过程中,产生3种类型的愈伤组织,其中呈紫红色的胚性愈伤组织芽分化最多.适宜愈伤组织诱导的培养基为:MS+6-BA2mg/L+IBA0.2mg/L,诱导率可达90.48%;适宜芽分化和继代增殖的培养基为MS+6-BA0.4mg/L+NAA0.05mg/L,继代培养40d,平均增殖倍数11.72倍;适宜生根的培养基为1/2MS+IAA0.2mg/L+活性炭0.5%;过渡移栽基质为腐殖土:河沙=2∶1,试管苗根长约1cm时移栽,保湿,成活率高达72%.
The callus was produced from the aseptic leaves of fresh, tender saussurea inuol uorata kar, etkir cultured on MS Media which contained different hormones for 15days. In the course of inducing callus, the purple-red was embryonic callus and bud differentiation was the most. The most suitable medium of callus induction was MS+6-BA2mg/L+IBA0.2 mg/L and the rate of induction reached to 90.48%; the bud differentiation and subculture medium was MS+6-BA0.4mg/L+NAA0.05mg/L, after subcultured 40days, the average propagation number was 11.72; the rooting medium was 1/2MS+IAA0.2mg/L+active carbon 0.5%. The transition substrate for transplant was humus soil: river sand=2:1. The rooted tube seedling transplanted and keep moisture when the root length was 1cm, and then the surviving rate highly reached 72%.
