摘要
以薯蓣皂甙元的收率为指标,考察了从豆科植物胡芦巴种子提取薯蓣皂甙元的水提和乙醇提两种工艺路线的优劣,并应用L9(34)正交试验设计优化了乙醇提工艺条件,得到以下结论:采用醇提取工艺有利于提高皂甙元的产率;影响乙醇提取工艺薯蓣皂甙元含量的主次顺序为:A>D>B>C(A乙醇浓度;B乙醇用量;C回流次;D回流时间),其最佳的工艺条件是:脱脂除胶胡芦巴豆粉50g,加入12倍于胡芦巴豆粉95%乙醇,加热回流提取3次,每次提取3h。
In the yield of diosgenin as a marker,the alcoholextraction and waterextraction technology about extracting diosgenin from fenugreek seeds were compared,the optimal condition of alcoholextraction were investigated by L(34)orthogonal design.The conclusion can be summarized as follows:the alcoholextraction technology was beneficial to increasing yield of diosgenin;and the sequence of factors that effects diosgenin yield in ethanol extraction is A>D>B>C(A:ethanol concentration; B:the amount of ethanol;C:refluxing number of times;D:refluxing time).The optimal condition obtained is that 50 g of degreased and degummed fenugreek seeds ware refluxed for 3 hours with 600 ml 95%ethanol and the refluxing number of times is 3.
