摘要
采用反相高效液相色谱法测定了发酵液中的纳他霉素。在Phenomenexprodigy5μODS3100A(250mm×4.60mmi.d.,5μm)色谱柱上,以甲醇 水 磷酸(体积比85∶15∶0 15)为流动相,体积流速为1mL/min,紫外检测波长为303nm。实验结果表明,该方法的相对标准偏差为0 24%(n=5),回收率为99%以上。
A method for determining natamycin in fermentation broth by RPHPLC is described.The operating conditions were Phenomenex prodigy 5μ ODS3 100A(250 mm×4.60 mm i.d.,5 μm)at room temperature,methanolwaterphosphoric acid(85∶15∶0.15,V/V)as mobile phase with a flow rate of 1.0 mL/min and UV detection at 303 nm.The retention time of natamycin was 1.972 min.This method is simple,rapid and accurate.The results will not be affected by other components in the broth.The relative standard deviation was 0.24%(n=5), and the recovery was over 99%.
