白腐菌产木素过氧化物酶发酵条件的优化 被引量:11

OPTIMUM CONDITIONS FOR PRODUCTION OF LIGNIN PEROSIDASE BY WHITE ROT FUNFUS

摘要 在静置和振荡两种培养条件下研究营养条件对白腐菌合成木素过氧化物酶(1igninperoxidase,LiP)的影响。静置培养时,LiP在碳氮比(C/N)低的培养基中显示较高的酶活力,碳源以葡萄糖(0.02%)和糊精(0.18%)同时存在及分段加入要比单一葡萄糖作为碳源时获得更高的酶活;振荡培养时,在碳氮比高的培养基中LiP酶活最高,而类似于静置培养的氮源组合及分段模式却明显地抑制了LiP的合成。优化后,静置培养在第6天可获得7560U/L的酶活;振荡培养在第8天获得6500U/L的酶活。发酵液中LiP酶活力与菌体分泌的其同功酶的数量成一定的对应关系。 This paper investigates effects of nutrient conditions on lignin peroxidase(LiP) activity in static and shaking culture. In static condition,LiP appears high activity in carbon limiting medium ,with glucose (0.02%) and dextrin(0.18%) together or batch adding as carbon sources LiP activity is much higher than with sole glucose;In shaking culture ,LiP appears high activity in nitrogen limiting medium,the similar nitrogen source combination and batch model inhibit LiP production. After majorization of medium ,LiP activity gets to 7560 U/L and suitable productive time is 6d in static condition;Under shaking condition ,LiP activity arrives 6500 U/L and suitable productive time is 8d. There is proportional relationship between LiP activity and numbers of LiP isoenzyme.
