摘要
根据未成熟的胸腺细胞对可的松类激素敏感的特性,给BALB/C小鼠腹腔注射地塞米松(DX,125mg/kg)。注射后12、24小时取胸腺,光镜和电镜观察胸腺悬液及组织切片内的胸腺哺育细胞(TNC)。DX使未成熟的胸腺细胞变性和碎解,细胞密度降低,光镜和电镜下观察切片,均可明确辨认TNC的存在。证明动物应用DX注射后,光镜下可简便地确认胸腺内的TNC。
Immature thymocytes are cortisone sensitive. According to this, BAIB/c mice were injected intraperitoneally with a single dose of dexamethasone (DX) (125mg/kg body weight).Following DX administration, mice were sacrificed at 12, 24 hours and their thymuses were removed for light and electron microscopic studies : 1) TNCs in vitro in thymic cells suspensions, and 2) epithelial cells contained thymocytes in situ in tissue sections. The results show that many of immature thymocytes were degradated or delepted. The tissue cell density were reduced. Not only underelectron but also under light microscope, a special kind degradated lymphoepithelial complexes can be seen in thymic cortex, while the changes of TNCs in thymic cell suspension are similar to that of the special lymphoepithelial cells in thymic cortex at the same time intervals.lt demonstrates simply that TNC are in situ in thymic cortex.
