摘要
本实验应用显微分光光度计分析了经SDS——EDTA处理后的人精液标本,用苯胺蓝染色显示赖氨酸型组蛋白,计量核蛋白相时含量。结果表明正常生育力男性的精子经SDS-EDTA处理后,苯胺绿染色阳性的精子数增多,且染色强度亦增大。经SDS-EDTA解聚后,80%的苯胺蓝染色阳性精子核中的组蛋白相对含量均>2.3AU,而未经SDS-EDTA处理的样本中,苯胺蓝阳性精子的组蛋白相对含量均<2.3AU。故本实验阐明了精子核蛋白经解聚后再作组蛋白含量测定较为正确。
The quantification of lysine-rich histone content of ejacuated sperms after SDS-EDTA treatment was conducted using miorospectrophotometry of aniline blue stained semen smear (to demonstrate lysine protein). It has been reported that the percentage and stainibility of aniline blue positive sperm increase after SDS-EDTA treatment. After decondensation by SDS-EDTA, the relative content of lysine proteins≥2.3 as compared with≤2.3 in untreated specimens. It has been suggested that the disulfide bonds in sperm nuclei could be broken under the action of SDS, providing more opportunities for binding between aniline blue and lysine residues of histones, and that the quantification of relative lysine-rich histone was more accurate after decondensation of ejaculated sperm.
