蜜环菌多糖Am -Ⅰ的部分理化性质及结构研究

摘要 液体培养的蜜环菌水溶性胞外粗多糖经纯化得Am-Ⅰ,再经Sephadex G-200柱层析和醋酸纤维薄膜电泳鉴定Am-Ⅰ为均一性组分。采用凝胶色谱法得Am-Ⅰ相对分子量约为6.65×105Dal,旋光法得[α]D25为+100°,苯酚-硫酸法、Folin-酚法、Dische反应法测其糖含量、蛋白质含量和糖醛酸含量分别为91.51%、4.78%和2.81%。气相色谱分析表明:Am-I的单糖组成为D-葡萄糖、D-岩藻糖、D-阿拉伯糖、D-甘露糖、D-鼠李糖、D半乳糖、摩尔比为:17.05:0.33:0.36:1:0.42:13.67。红外光谱、纸色谱、Smith降解分析表明:Am-I是一种含半乳糖醛酸的酸性多糖,其结构主要由吡喃型单糖通过β(1→4)键和β(1→6)键连接而成。β-消去反应表明这种糖蛋白是非O-型糖肽键。 Am-Ⅰ was isolated from the Armillaria mellea and purified by DEAE-cellulose and Sephadex G-200 columnchromatography. The following results were obtained: Am-Ⅰwas an acid polysaccharide with galacturonic acid. The molecularweight of Am-Ⅰwas 6.65×105Dal. The specific spin power of the polysaccharides was Am-Ⅰ+100°. Its carbohydrate contentwas 91.51%. The urinic acid and protein content of Am-Ⅰwere respectively 2.81% and 4.78%. The gas chromatography(GC)indicated that Am-Ⅰwas composed of D-glucose, D-fucose, D-arabinose, D-mannose, D-rhamnose, D-galactose respectivelyin the molar ratio of 17.05:0.33:0.36:1:0.42:13.67. The IR spectrum of Am-Ⅰshowed a character of β-glycosidic bond in 400~4000cm-1. Smith degradation showed that the main strand of Am-Ⅰconsisted of monosaccharides which were linked by β(1,4) andβ(1,6) glycosidic bond. The glycopeptidic bond of Am-Ⅰwas non-O-type one.
