H-FABP基因型对二花脸猪相关性状影响的初步分析 被引量:21

摘要 利用21窝不同日龄的二花脸猪共97头,根据日龄分布,分成1组(日龄阶段)—49天,2组—58~60天,3组—61~70天,4组—71~80天,5组—81~90天,6组—228天六个日龄组;采用索氏脂肪抽提法和石蜡组织切片图像分析法对其背最长肌及半腱肌的肌肉组织学特性进行了测定。根据心脏脂肪酸结合蛋白(HeartFatty Acid Binding Protein,H FABP)基因的PCR RFLP多态性,对其与二花脸猪相关性状初生重、20日龄体重、断奶重及肌肉组织学特性的关系进行了分析。所有二花脸猪均表现为AADD型。邓肯氏(Dancans)差异显著性检验结果表明:(1)二花脸猪H FABP基因HH与Hh、hh基因型间20日龄体重差异显著(P<0 05),但对初生重及断奶重无显著影响;三性状值均表现为HH>Hh>hh。(2)在不同的基因型间,没有一个日龄组内的肌肉组织学性状值表现为差异显著,从侯选基因角度分析H FABP基因对肌肉粗脂肪含量的影响,发现趋向于HH>Hh>hh。 In this study,ninetyseven Erhualian pig from 21 litters were utilized and divided into six groups according to their day age distribution,that is,group one49 d,group two58~60 d,group three61~70 d,group four71~80 d,group five81~90 d,group six228 d.The m.longissimus over the last second to third vertebra thoracica and the middle m.semitendinosus were taken out as samples for muscular histological study by tissue paraffin sectionimage analysis method.And the m.logissimus over the second to fifth vetebra lumbalis were taken out for intramuscular fat content measurement by Soxhlet's method.Based on HFABP genetic variation detection,the effect of HFABP gene on birth weight,20day weight,weaning weight and muscular histological characteristics in Chinese Erhualian pig were analyzed.All the Chinese Erhualian pigs genotyped as AADD.The results of Dancans analysis of variance indicated that significant variance was found on 20day weight(P<005) between HH and Hh,hh genotype animals.But no significant difference was found between various genotypes on birth weight and weaning weight although all traits values tended to be in a formula of HH>Hh>hh.No significant variance was found in all muscular histological characteristics among HH,Hh,hh genotype animals in all groups.Considered as a candidate gene for IMF,the effect of HFABP gene on IMF in the dried m.longissimus tended to be HH>Hh>hh.
