摘要
目的 探讨牛磺酸对过氧化氢所诱发的晶状体上皮细胞凋亡的抑制作用。方法取兔晶状体离体培养,分为对照组、H_2O_2氧化损伤组、氧化损伤同时加牛磺酸共3组,每组64只晶状体,分别以不同时限观察各组晶状体混浊情况,DNA缺口末端原位检测法(TUNEL法)及DNA片段分析检测各组晶状体上皮细胞凋亡情况。结果 牛磺酸组的晶状体混浊情况和晶状体上皮细胞凋亡率均明显低于H_2O_2氧化损伤组,与对照组差别无统计学意义。DNA片段琼脂糖凝胶电泳:H_2O_2组培养24、36、48及72h各时限均呈现凋亡细胞的典型梯状条带;而培养24h的对照组、牛磺酸组均未出现此变化而仅呈现正常细胞电泳条带。结论 牛磺酸可抑制H_2O_2氧化损伤所诱导的兔晶状体上皮细胞凋亡,从而延缓或减轻白内障形成。
Objective To investigate the inhibition effect of antioxidants (taurine) on the lens epithelial cell apoptosis induced by H2O2. Methods Rabbits lenses in vitro were incubated in the minimum essential medium (MEM), and were divided into three groups: control group, H2O2 group and H2O2 added taurine group. The opacity of the lenses was detected. The DNA fragmentation assay of lens epithelial cells and TUNEL staining were examined. Results The opacity and the apoptosis rate of the taurine group was apparently lower than the H2O2 group. By DNA fragmentation assay, the DNA ladder was found in H2O2 group during 24-72 hours, while DNA ladder was not found in normal group and H2O2 added taurine group. Conclusion Lens can take refuge from the oxidative injury of H2O2 with the addition of taurine, as a result, the apoptosis rate of lens epithelial cells can be restrained and cataract formation alleviated.
