SPR生化分析仪对6-氧甲基鸟嘌呤-DNA甲基转移酶的检测 被引量:16

摘要 6-氧甲基鸟嘌吟-DNA甲基转移酶(简称MGMT)是一种重要的DNA损伤修复酶,组织中MGMT活性水平的检测可以为肿瘤的早期预防和治疗中化疗方案的确定提供依据。我们利用基于表面等离子体谐振(SPR)生物传感器原理研制成的SPR-2000型生化分析仪对MGMT的基因片段进行了检测,然后采用配套的软件系统对实验数据进行处理,获得检测曲线;再对检测曲线进行温度补偿修正后便获得了杂交反应曲线,SPP-2000型生化分析仪可以检测1.18×10^(-10)mol/L浓度的MGMT核酸样品,为进一步进行临床检测的研究奠定了基础。
