摘要
采用马铃薯Y病毒属通用引物,从甘薯羽状斑驳病毒(SPFMV)山东济南分离物和浙江杭州分离物基因组3′ 末端扩增到一个含1814个核苷酸的序列,序列号分别为AJ310201和AJ310202。两个序列均包括NIb基因部分序列(nts1 645)、外壳蛋白基因(nts646 1590)和3′ UTR(nts1594-1814),3′ 末端具有典型的poly(A)尾。外壳蛋白基因核苷酸系统进化树分析和氨基酸分析表明,以前对一些SPFMV分离物的命名有待商榷,一些相关性较强的分离物可分为3个进化主要群体,我国两个分离物属于第二群体。
Using a pair of universal primers of family Potyviridae,a DNA fragment of 1814 nucleotides were amplified, cloned and sequenced from 3′terminal part of Sweet potato feathery mottle virus(SPFMV) from Jinan and Hangzhou isolates .Both of them were contained part of NIb gene(nts 1645),CP gene(nts 6461590) , 3′UTR (nts 15941814) and typical poly(A) tail. Analysis of the nucleotides and amino acid phylogenetic tree of coat protein genes showed that the previously identification of some virus isolates were wrong, and some related isolates were classed to be three evolutional groups. The isolates of Jinan and Hangzhou belonged to group II.
