摘要
目的 :探讨HPV 6bL1VLPs经三种途径免疫小鼠后的免疫反应 ,以选择机体获得免疫保护的最佳途径。方法 :小鼠以鼻腔吸入、阴道免疫、肌肉注射HPV 6bL1VLPs分为三组 ,经间隔 2w三次免疫后取血、肺冲洗液和阴道冲洗液标本 ,用ELISA法检测血清IgG和分泌物IgA抗体 ,并用血凝抑制试验检测抗体的中和作用。结果 :HPV 6bL1VLPs经肌肉注射、鼻腔吸入均能诱导产生血清IgG和呼吸道、阴道粘膜的IgA抗体 ,经阴道免疫可产生血清IgG和阴道粘膜的IgA抗体 ;抗体均具有中和活性 ;其中肌肉注射组产生的血清IgG抗体滴度最高 ,鼻腔免疫产生的粘膜IgA抗体滴度最高。结论 :HPV 6bL1VLPs具较强抗原性 ;肌肉注射和鼻腔吸入免疫可刺激小鼠产生全身和局部粘膜的中和抗体 。
Objective:To study the immune response to HPV6b L1 VLPs in mice and explore the best route of immunization against HPV infection.Methods:HPV6b L1 VLPs were used to immunize mice by three different routes of intramuscular,intranasal and vaginal immunization. After three immunization,the samples of serum,lungs washes and vaginal washes were taken from the mice simultaneously. HPV6b L1 VLPs specific serum IgG and mucosal secretory IgA antibodies were tested by ELISA and the neutrolizing effects were tested with hemagglutination inhibition assay.Results:Specific serum IgG and secretory IgA with neutralizing activity in trachea and vagina could both be induced by intramuscular and intranasal routes,but vaginal immunization could only produce serum IgG and secretory IgA in vaginal. In particular,the highest titer of serum IgG could be induced by intramuscular immunization,and the highest titer of secretory IgA in trachea and vagina could be induced by intranasal immunization.Conclusion:HPV6b L1 VLPs have strong immunogenicity and can elicit significant titers of neutralizing antibodies in sera and vaginal secretions in mice by intramuscular or intranasal immunization .Intranasal immunization may be the best route to induce immmune response.
