实验性暴发性肝衰竭大鼠肝脏NK细胞与肝再生的关系 被引量:3

Relationship Between Liver Resident NK Cells and Liver Regeneration in Experimental Fulminant Hepatic Failure

摘要 目的 :探讨实验性暴发性肝衰竭时大鼠肝脏自然杀伤 (NK)细胞与肝再生的关系。方法 :制备实验性暴发性肝衰竭大鼠模型 ,采用免疫组织化学染色技术分析增殖细胞核抗原 (PCNA)阳性细胞百分率 ,采用乳酸脱氢酶释放试验计算NK细胞活性。结果 :实验组肝细胞PCNA表达占 (9.2 0± 2 .6 4 ) % ,低于对照组 (2 6 .6 5± 3.91) % ,P<0 .0 0 1;实验组肝脏NK细胞活性 (16 .38± 3.32 ) % ,高于对照组 (11.70± 3.17) % ,P<0 .0 0 1。实验组肝细胞PCNA表达百分率与实验组NK细胞活性之间呈较强的负相关。结论 :实验性暴发性肝衰竭大鼠肝再生不充分 ,肝脏NK细胞活性增加 ,肝再生与NK细胞活性呈较强的负相关。 Objective: Our aim was to study the relationship of liver resident natural killer(NK) cells and liver regeneration in experimental fulminant hepatic failure(FHF). Methods:Models of FHF(rats) were induced by lipopolysaccharide. Expression of proliferating cell nuclear antigen(PCNA) was observed with techniques of immunohistochemistry. Activity of NK cells was detected by the tests of release of lactate dehydrogenase(LDH). Results:The positive rate of PCNA expression in hepatocytes in the rats with FHF was lower than that in the control group( 9.20%± 2.64% vs 26.65%±3.91%,P<0.001). The activity of liver resident NK cells in rats with FHF was higher than that in the control group( 16.38%±3.32% vs 11.70%±3.17%, P<0.001). No relation between liver resident NK activity and positive rate of PCNA expression was found. Conclusion:Liver resident NK cells may be involved in the impairment liver regeneration in rats with FHF.
